AAV production processes can be challenging to standardize, making process robustness a critical consideration as developers work toward scalable manufacturing. In collaboration with Thermo Fisher Scientific, Culture Biosciences evaluated the robustness of the Gibco™ AAV-MAX Helper-Free AAV Production System in cloud-native 250 mL bioreactors.

By systematically evaluating key process parameters, including dissolved oxygen, agitation, pH at transfection, and viable cell density, the study demonstrated comparable viral genome titers and full capsid percentages across the tested ranges.
